What challenges shape Pharma?

Contamination control strategy per Annex 1

The EU GMP Guide Annex 1, published in 2022 and applicable since August 2023, requires a cohesively documented contamination control strategy (CCS) for sterile manufacturing that ties together facility design, personnel flow, environmental monitoring, and barrier technology. Many sites have individual measures in place, but no robust CCS as a coherent body of evidence.

Inspection readiness for FDA and EMA

FDA inspections per 21 CFR Part 210/211 and EMA inspections per EudraLex Volume 4 end, where deficiencies are found, in Form 483 observations, warning letters, or a statement of non-compliance. Anyone who only establishes audit readiness shortly before the date is repairing under time pressure rather than during ongoing operations, and risks recurring findings.

Data integrity and computer system validation

FDA and EMA assess electronic records against 21 CFR Part 11 and the EU GMP Guide Annex 11 along the ALCOA+ principles. Shared logins, unreviewed audit trails, and raw data that can be overwritten after the fact are among the most common data integrity findings in computerized GxP systems.

Tech transfer and process validation

The transfer from development to commercial scale requires qualification and validation per EU GMP Guide Annex 15 as well as a process performance qualification, underpinned by ICH Q8 to Q10. If the knowledge from development is not handed over cleanly, validation fails on parameters whose rationale is missing at the new scale.

GDP-compliant distribution of temperature-sensitive medicinal products

The EU GDP Guidelines 2013/C 343/01 require that the quality achieved in manufacturing is preserved all the way to the recipient. For cold-chain biologics and vaccines, temperature mapping, continuous monitoring, and qualified transport routes become a core operational requirement, not a logistics afterthought.

In pharma it is rarely the single SOP that decides the outcome, but rather the handover between development, production, quality and logistics. Teams that treat data integrity, process validation and inspection readiness as one from the start pass the inspection because operations already live it.

The 2022 version of the EU GMP Guide Annex 1 requires a documented contamination control strategy that brings together all contamination-relevant elements of a site: facility and cleanroom design, personnel and material flow, environmental monitoring, barrier and isolator technology, and the underlying quality risk management per ICH Q9. What matters is not the sum of the individual measures, but the evidence that they work as a coherent concept.
